Effect of Rodenticide Vacor Poisoning on Insulin Binding to Adipocytes and Erythrocytes of Rats

Abstract
Vacor, when administered orally at a small dose (5mg/kg body weight) to rats, caused a glucose intolerance without apparent fasting hyperglycemia. At the same time, Vacor caused a reduction of insulin binding to epididymal adipocytes of rats. Scatchard analysis revealed that the reduction of adipocyte insulin binding in Vacor-poisoned rats is mainly accounted for by a decrease in the insulin receptor concentrations. The erythrocyte insulin binding, however, was not affected by Vacor-poisoning.
These results suggest that Vacor-induced decrease in the insulin receptor concentrations of peripheral tissues such as adipocytes causes a reduced tissue response to insulin and eventually leads to glucose intolerance in Vacor-poisoned rats.
